INTRODUCTION Utilisation of elements to relieve man and animals from disorders, as well as for maintenance of healthy status, is as old as life on earth. The Ayurvedic medicinal system has been in vogue since the Vedic period or even earlier. It emphasizes on the maintenance, promotion of health and curing the diseases. Thus Ayurveda was developed to safe guard Arogya (health) which is considered to be essential for the achievement of the four primary objects of life viz. Dharma, Artha, Kama and Moksha. For achievement of these goals, medicines especially Rasaushdhis have an important role. Rasa Chikitsa is the best therapy among others due to quicker recovery from diseases and effective in even very small doses. In present era Ayurvedic physicians profusely use minerals, metals, gems and some of animals as well as vegetable products. While our modern counterparts rarely accept metallic preparation for internal use, as modern pharmacology does not recommend such type of preparation for therapeutic purposes. Ayurvedic physicians are of the view that different Ayurvedic pharmaceutical processing’s like Shodhana, Marana, Samskaras; Jarana etc. convert metallic preparations into non-toxic. The standards of Ayurvedic medicines available in the market are always questioned. The general belief that Ayurvedic drugs are harmless and have zero toxicity is dangerous. Rasaushdhis especially if they are not prepared by proper method may turn out very harmful. Very minute fault in preparation of Rasaushdhis can produce major complication in the patient. The present work taken up for the study entitled “Pharmaco-clinical study on malla sindoora and its effect in the management of amavata”. The need for the study is highlighted here. Rasa shashtra one of the pharmaco-therapeutic branch, utilizes mercury in three forms viz… Murchita, Marita and baddha. In murchana, parada after purification treated with many other drugs to get disease curing property in it. Using this principle as a platform the most superior, unique formation of rasa shastra, kupipakwa rasayana was developed. Malla sindoor one of type of sagandha, bhirdhooma kupipakwa rasayana claimed as vata-kapha hara and indicated in sarva-vata rogas. In this present research work we have selected malla sindoora as our test drug on virtue of its vital importance in therapeutics as well as particular skill involved in its pharmaceutical preparation. In this study malla sindoora was prepared according to the guideline of siddha bhesajaya manimala. Keeping therapeutic indication mentioned in our classics one may conclude that malla sindoora is effective in disorders with dominance of vata and kapha so we had decided to perform an experimental study to find out the effect of the same in locomotor system on suitable models. Amavata is one such disease which is mainly caused due to the disarrangement of agni, precisely jatharagni results in the production of ama and this ama circulates in the whole body propelled by vitiated vata dosha producing blocks in the body channels giving rise to amavata. This is being correlated to rheumatoid arthritis on the basis of etiology, pathology, therapeutics, signs and symptoms. Now rheumatoid arthritis is the greatest crippling disease of unknown causation, claiming the maximum loss of human working power and was second amongst the disease which produced chronic limitation to major activities. The heart disease is being the first. An enormous amount of research work has been done so far on this problem all over the world; some effective drugs have also been searched. But none of them can be considered as specific. The whole work has been categorized in to 1. Introduction: Explains the need of the study and hypothesis behind the study. 2. Review of literature: Deals with drug review and disease review comprising of both modern and ayurvedic aspects. 3. Pharmaceutical study: Includes detailed documentation of the preparation of Mallasindoora carried out. 4. Analytical study: This part includes the physio- chemical analysis and Namburi phased spot test of Rasakarpoora and Mallasindoora. 5. Clinical study: Includes observational study on Mallasindoora in the cases of Amavata. 6. Discussion: This part includes observations, findings and results of various studies and possible explanation. 7. Conclusion & Summary: In this part conclusion drawn is presented with summary of whole study.
