Group 4 Project Biology:
Biology class when participating in group 4 project tested chromatography TLC and level of photosynthesis in elodea plants in different light.
CHROMATOGRAPHY Extraction and separation of plant pigments.
The aim of our group 4 project was extraction of plant pigments chlorophyll from Elodea plant and (red plant) and chromatographic separation of the mixtures by using a thin layer chromatography.

The value of Rf is defined as the ratio of the distance X (virtually) (equal to the displacement of the center spot from where you started) to the distance Y (equal to postpone the solvent front):
However the extraction of pigments from the leaves of the two plants were done by the chemistry group, however in our biology class as Alex is in chemistry we took elodea plant with red leaves bur green pigment and Alex heated it up with ethanol for an hour, I was watching it as unfortunately I do not take chemistry but was interested how Alex conducted it and what happened to the plant during and after the heating up.

Then after some time the extraction of the pigments (the green liquid) was used for TLC.
* Alex had drawed the 1cm on side and 1 cm from the other on a piece of TLC paper. And then the present people were split into two groups, I was with Alex and we took the pipet and droped 2 drops of the solution and placed It into the jar, while at the same time during all of the group 4 project I was taking pictures.
* Then once the results of TLC chromatography were visible we took out the papers carefully not to touch them on the surface with fingers and I with alex measured the results, (Rf)

LEVEL OF PHOTOSYNTHESIS IN ELODEA PLANT IN DIFFERENT.
The boxes were prepared and equipped all prepared by the chemistry group and by the physics group, the final results we received the boxes all prepared to be used.
What was done:
* I took part in helping the girls find the needed plant, and it was then purchased.
* I then with Alexandra arranged the biology group giving instructions who done what. The class was arranged in to 5 groups each undertaking a certain box with a certain colour, mine and Alex’s box was natural light. After the students knew what their box was, I wrote on the board with the help of class to what has to be measured, and made sure everyone knew how to measure it.

* The Elodea plant was taken and cut with a ruler in to 5cm ( by other girls) whereas me and Alex took the challenge our self’s
Firstly we measured the ph of the mixed water being tap water and mineral, and then placed the elodea in 7 test tubes, for each group meaning 70 test tubes al together

* Then each group filled their testubes with 15 ml of water which was a mixture of tap & mineral water. Me and Alex when finished our own filling the testubes with the solution went around to make sure that everybody knew what they were doing, and I personaly gave a helping hand to one of the group ( Sandra and Natalia) who were a bit behind because they were not completely sure what we had to do and why. After all was sorted out out
* Then the each test tube was placed in a beaker, and sealed with a see through foil and me and Paula quickly cut up the foil for everyone just to prevent making a mess, then..
* I had to make sure that everyone knew what the next step was, and made sure that everyone at absolutely at the same time put the beaker with 7 test tubes inside the box, but just before I and Alex carefully plucked the electricity and made sure that every box was fully working and ready to have the beakers put in.
* I arranged a little practice just before to make sure that the people putting in the boxes the beakers with test tubes were able to close and open the box quickly enough, in order to maintain equality when put it, this was done in order just to give a clue how to do it because some students were rather on their own moon or weren’t interested, but as most of the group were and wanted the results to be good and with our errors.
* We then on the count of 3 put in the beakers with test tubes into the boxes and waited for a whole 7 days.
7 DAYS AFTER:

After we came back we took out the beakers with test tubes and yet again gave instructions to what had to be measured:
* Then I took out my permanent marker and numbered the test tubes from 1-7 very quickly in order so that when recording the data each group knew which test tube and could not make an error in measuring.
* Then I and Alex put the representative of each groups together to see what group had the most bubbles: ( I noted it down on the board)
* -After Me and Alex and the other groups; we used a piped and for every test tube recorded the volume of water, alex sucked up the water and measure while I recorded the result and poured the water into a main beaker where later after all the water was collected pH was measured.
* Then when the results were taken down, all the water was in one beaker, then me and Alex measured the pH of the water.
* Then when we finished I and Alex went around to see if others needed help, and after when everyone was finished I had to make sure each had worked out their average volume, and pH and for one group who were not capable of doing it I and Alex worked it out as well as then Alex called out the results of each group while I dropped it down on the board.
* Then after all was done we discussed that some groups made major errors when measuring but unfortunately we could not be responsible for others, we tried our best with Alex and Karolina to assure everyone was doing it correct but we didn’t manage fully. This is a shame really.
* Then after the class as soon as the bell rang everyone escaped and was only me , Alex and the teacher left to clean up the mess made by our class, no one bothered to help.
