Fluorescence Microscope, Cellular Organelles, and the Cytoskeleton
ABSTRACT:
Before the lab began, we started by learning the fluorescent microscope parts to be used, by examining the diagram given to us. Then, learning the theories and key concepts of the lab such as photo bleaching and stokes shift. Once we felt comfortable with the florescent microscope we created slides using dog kidney cells, onion cells, human cheek cells, antibody tubulin, and damaged elodea cells. These cells were stained with compounds such as, Alexa 488 ConA lectin, DAPI, and PI or Propidium iodide. The purpose of the stains is to show a few cellular organelle components. We were able to look at cell proteins such as tubulin; Tubulin was our protein of interest that was localized by the antibody from either rabbits or goats. The class was placed into groups and each group used a number of cells variously stained, this way we could divide up labor and view more of the cellular components. The cells were then copied from the monitor and printed for future viewing.
